Chromatin organizes nuclear DNA, but whether it actively regulates whole-cell mechanics remains unknown. Here, we used NETosis to show that chromatin decompaction within the nucleus increases plasma membrane tension and cell volume, establishing chromatin as a regulator of cell mechanics.
