From identifying individual protein-protein interactions (PPIs) in the 1980s to characterizing whole-cell interactomes more recently, approaches for identifying cellular PPIs have opened the way for understanding how protein complexes and molecular machines operate. These data, coupled with structural and computational techniques, offer functional insights and uncover how interactions are perturbed in pathogenic contexts.
