The exact routes and mechanisms of iron acquisition by mitochondria are unclear. We identify mitochondria–endoplasmic reticulum contact sites as an iron supply hub and reveal that the mitochondrial ubiquitin ligase MITOL regulates HMOX2 activity, to enable delivery of haem-derived iron to sustain mitochondrial iron homeostasis.
