The mitochondrial–peroxisomal fission adaptor MFF is identified as a selective pro-ferroptotic mediator, and the newly developed MFF–SPARK sensor enables high-content screening of ferroptosis-sensitizing compounds and the discovery of upstream regulators of MFF.
