Ten-year APHINITY data show Roche’s Perjeta-based regimen reduced the risk of death by 17% in people with HER2-positive early-stage breast cancer

The APHINITY study has demonstrated a clinically meaningful survival benefit when adding adjuvant Perjeta to Herceptin and chemotherapy in people with HER2-positive early-stage breast cancer. After ten years, the risk of death was reduced by 17% for people treated with the Perjeta-based regimen. The study showed a 21% reduction in the risk of death in the prespecified subgroup of people with lymph node-positive disease. The Perjeta-based regimen is approved in over 120 countries for the treatment of both early-stage and metastatic HER2-positive breast cancer. The regimen has been shown to almost double the rate of pathological complete response compared to Herceptin and chemotherapy in the neoadjuvant setting. The combination has also been shown to significantly reduce the risk of recurrence of invasive disease or death in the adjuvant setting. The full results of the APHINITY study will be presented as a late-breaking abstract at the 2025 European Society for Medical Oncology Breast Cancer Congress. The study's findings validate the sustained benefits of the Perjeta-based regimen as a well-established standard-of-care treatment in the curative setting. The safety profile of the Perjeta-based regimen was consistent with previous studies, and no new or unexpected safety signals were identified.
