This review covers the biology of the cGAS-STING pathway, the cell’s alarm system that shapes immune and stress responses with impacts on a range of processes, from antiviral defense and tumor immunity to chronic inflammation and disease. The underlying biology of cGAS and STING signaling will guide new therapeutics.
